鏈霉蛋白酶消化酶的配制
貯存液濃度20mg／m1，-20℃保存。*佳反應緩沖液為0．01mol／LTris—HCl(pH7．8)，0.01mol／LEDTA和0．5％SDS。常用工作濃度lmg／m1，工作溫度37℃。
消化酶的選擇
在選擇用何種酶消化時，應針對要顯示的抗原成分不同而選用不同的酶。一般胃蛋白酶和菠蘿蛋白酶消化主要用于細胞間質抗原的檢測，例如纖連蛋白(fibronectin)、層黏連蛋白(1aminin，LN)和各型膠原的顯示；其余酶消化均可用于細胞內抗原的檢測。在配制蛋白酶時，應考慮到酶的*佳pH。例如胰蛋白酶的*佳pH為7．6，要在鈣離子的激活下于37℃才能達到*佳效率；而胃蛋白酶則在酸性pH值下才能達到*佳效率。另外，經酶消化后的切片應充分洗滌，以去除殘留的消化酶，否則會對抗體或組織進行緩慢消化，影響抗原抗體的結合，破壞組織結構。陽性和陰性對照片應同時消化，避免人為差異。

Specificity: Mixture of several nonspecific endo- and exoproteases that generally digest proteins down to single amino acids. Resolves carboxylic acids and alcohols.
Working concentration: 0.5 - 2 mg/ml
