　　上海玉博生物——大鼠視網(wǎng)膜色素上皮細(xì)胞|細(xì)胞系|細(xì)胞株|腫瘤細(xì)胞，公司不僅有大量的**細(xì)胞，還提供大鼠視網(wǎng)膜色素上皮細(xì)胞的全程后期服務(wù)，可以向?qū)I(yè)人士咨詢?cè)敿?xì)的大鼠視網(wǎng)膜色素上皮細(xì)胞|細(xì)胞培養(yǎng)條件，凍存方法，及相關(guān)培養(yǎng)技術(shù)。

　　對(duì)于貼壁細(xì)胞，傳代可參考以下方法：

　　1. 棄去培養(yǎng)上清，用不含鈣、鎂離子的PBS潤(rùn)洗細(xì)胞1-2次。

　　2. 加2ml消化液(0.25%Trypsin-0.53mM EDTA)于培養(yǎng)瓶中，置于37℃培養(yǎng)箱中消化1-2分鐘，然后在顯微鏡下觀察大鼠視網(wǎng)膜色素上皮細(xì)胞|細(xì)胞消化情況，若細(xì)胞大部分變圓并脫落，迅速拿回操作臺(tái)，輕敲幾下培養(yǎng)瓶后加少量培養(yǎng)基終止消化。

　　3. 按6-8ml/瓶補(bǔ)加培養(yǎng)基，輕輕打勻后吸出，在1000RPM條件下離心4分鐘，棄去上清液，補(bǔ)加1-2mL培養(yǎng)液后吹勻。

　　4. 將細(xì)胞懸液按1：2到1：5的比例分到新的含8ml培養(yǎng)基的新皿中或者瓶中。

　　對(duì)于懸浮細(xì)胞，傳代可參考以下方法：

　　方法一：收集細(xì)胞，1000RPM條件下離心4分鐘，棄去上清液，補(bǔ)加1-2mL培養(yǎng)液后吹勻，將細(xì)胞懸液按1：2到1：5的比例分到新的含8ml培養(yǎng)基的新皿中或者瓶中。

　　方法二：可選擇半數(shù)換液方式，棄去半數(shù)培養(yǎng)基后，將剩余細(xì)胞懸起，將細(xì)胞懸液按1：2到1：3的比例分到新的含8ml培養(yǎng)基的新皿中或者瓶中。

　　3)細(xì)胞凍存：待細(xì)胞生長(zhǎng)狀態(tài)良好時(shí)，可進(jìn)行大鼠視網(wǎng)膜色素上皮細(xì)胞|細(xì)胞凍存。貼壁細(xì)胞凍存時(shí)，棄去培養(yǎng)基后加入少量胰酶，細(xì)胞變圓脫落后，加入約1ml含血清的培養(yǎng)基后加入凍存管中，再添加10%DMSO后進(jìn)行凍存。懸浮細(xì)胞凍存時(shí)，應(yīng)將細(xì)胞收集，1000RPM條件下離心4分鐘，少量保存上清液(防止細(xì)胞吸走)，加入部分新鮮培養(yǎng)基，加入到凍存管中，在凍存管中加入10%DMSO后進(jìn)行凍存。
